Biphasic effect of xenon on norepinephrine (NE) release were observed in spontaneously beating guinea pig isolated atria. In the presence of 2 mM potassium-rich Tyrode solution (69 mM NaCl, 2.7 mM KCl, 3 mM CaCl2, 1.8 mM MgCl2, 10 mM HEPES, and 12 mM glucose, pH 7.4), noradrenergic nerve stimulation induced NE release. Xenon (0.5-2%, 0.4 min of exposure) inhibited both early and late phases of NE release. Ethanol (1%) significantly inhibited both phases of NE release. At a concentration of 2%, xenon completely inhibited NE release. The inhibitory effect of xenon was antagonized by sodium bicarbonate. The presence of Ca2+ or Ba2+ in the Tyrode solution suppressed the inhibitory action of xenon. In the presence of 1 mM EGTA, neither xenon nor ethanol inhibited NE release. We conclude that xenon inhibits both phases of NE release in the guinea pig isolated atria.
